實驗材料
0.4 % w/v trypan blue (GibcoBRL 15250-061)
Erythosin bluish stain
取0.1 gram Erythrosin bluish (Sigma E-9259) 及0.05 gram preservative methyl paraben (Sigma H-3647) 溶于100 ml Ca /Mg free saline
血球計數(shù)盤及蓋玻片(Hemocytometer and coverslip)

實驗步驟
1 取50ml 細胞懸浮液與50ml trypan blue (or Erythrosin bluish) 等體積混合均勻于1.5ml 小離心管中。
2 取少許混合液(約15ml) 自血球計數(shù)盤chamber 上方凹槽加入，蓋上蓋玻片，于100 倍倒立顯微鏡下觀察，活細胞不染色，死細胞則為藍色(或
紅色- Erythrosin bluish)。

3 計數(shù)四個大方格之細胞總數(shù)，再除4，乘以稀釋倍數(shù)(至少乘以2，因與trypan blue等體積混合)，*后乘以104 ，即為每ml 中細胞懸浮液之細
胞數(shù)。若細胞位于線上，只計上線與右線之細胞(或計下線與左線之細胞)。

4 大格*細胞總數(shù)x 2 x 104 / 4= 細胞數(shù)/ml*每一大格的體積= 0.1cm x 0.1cm x 0.01cm = 10-4 ml
4 若不用血球計數(shù)盤，可用Coulter counter 作自動計數(shù)，惟無法辨別死細胞或活細胞。
5 范例：T75 monolayer culture 制成10ml 細胞懸浮液，取0.1 ml 溶液與0.1ml trypan blue 混合均勻于試管中，取少許混合液加入血球計數(shù)盤，計數(shù)四大方格內(nèi)之細胞數(shù)目。
